We present a highly sensitive library construction protocol suitable for subcellular or ultralow input RNA sequencing (ulRNA-seq). In order to improve the sensitivity and low abundance gene detection ability of the library construction protocol, the key components of the library construction protocol, such as reverse transcriptase (RT), template-switching oligos (TSO) terminal modification and template RNA structure were optimized.
