Determination of medium composition to improve material production of transgenic E. coli

In this study, we use two transgenic Escherichia coli that produce different substances, 4APhe and Tyrosine, as models to optimize the medium. By comparing the two strains, we will consider the effect of different genotypes on substance production. As an experiment, first, the medium components of a single compound are mixed at various concentrations to prepare a comprehensive medium composition. Next, proliferation data and production volume data are acquired by plate reader culture and HPLC, and the relationship between cell proliferation and substance production is considered. After that, the composition of the optimized medium is determined by identifying the medium component that most contributes to the amount of substance production using decision tree learning and optimizing the concentration of the deciding component. A demonstration experiment will be conducted with the optimized medium composition, and a series of optimization methods and the optimized medium will be evaluated. Finally, we analyzed transcriptome to explain the increased production of 4APhe and Tyrosine.
