TET2 and DNMT3A regulate the sensitivity to PARP inhibitor Overall design: To model the somatic loss-of-function mutations found in human blood diseases, mice with conditional inactivation of Dnmt3a and Tet2 in the hematopoietic system were generated and further crossed with mice harboring Flt3ITD mutation. Lin-cKit+ bone marrow cells (BMC) were extracted from bone marrow of mutant and control mice. Extracted BMC were treated with ruxolitinib.
