Paired-end sequencing of PCR amplicons templated off of an omega-dSpCas9 expression plasmid library used for PAM-dependent B1H selection experiments in E. coli. The omega-dSpCas9 plasmid library harbored a NNS-randomized region spanning five consecutive codons (1332-1336) within the PAM-interacting domain of omega-dSpCas9; read pairs captured the entire NNS-randomized region on both strands. Amplicons were derived from post-selection library samples, each corresponding to one of two biological replicates tested for each selection configuration.
