Temozolomide (TMZ) resistance may contribute to the treatment failure in patients with glioblastoma (GBM). Hence, understanding the underlying mechanisms and developing effective strategies against TMZ resistance are highly desired in the clinic. long non-coding RNAs (lncRNAs) have emerged as new regulatory molecules with diverse functions in biological processes and been deregulated in many pathologies, involved in the therapeutic resistance. It is urgent to elucidate the underlying lncRNA-based mechanisms of TMZ resistance in GBM patients. Overall design: LN229 cells were seeded in 96 well plates with 6000 per hole and the half maximal inhibitory concentration (IC50) of TMZ was determined. Then, TMZ was added to the cell culture medium with IC50 1/50 concentration (0.83±0.26μM) to culture LN229 cells in 6 well plates. After cells grew stably, the drug dose was increased in multiples. Each dose was maintained for 15 days to the end of the fifth month. Induced TMZ-resistant cells were named as 229R. LN229 and 229R cells were used for lncRNA microarray
