Chitinase 1: A novel therapeutic target in metabolic dysfunction-associated steatohepatitis

Macrophage profiling was performed using single-nuclei RNA sequencing and flow cytometry. Single-nuclei RNA sequencing demonstrated a notable increase in macrophages numbers, particularly of lipid-associated macrophages, in MASH mice. Overall design: We established a mouse model of MASH-related liver fibrosis by administering streptozocin (STZ, 0.2 mg) in combination with a high-fat and high-cholesterol (HFHC) diet. Single-nuclei RNA sequencing (sNuc-Seq) was conducted using frozen liver tissue samples from one normal male C57BL/6 J mouse and two mice with MASH-related liver fibrosis.
