Transcriptome signatures after 5-AZA-dc and TSA stimulation of equine chondrocytes

Using the stimulation of passaged equine chondrocytes in monolayer culture with the 5-AZA-dc and TSA epigenetic modifiers we have investigated the state of chondrogenic differentiation of expanded cells on the transcriptome level. The results of next generation sequencing of 3 prime mRNAseq libraries on stimulated and unstimulated chondrocytes of the 3rd passage showed no significant influence of 5-AZA-dc treatment. Chondrocytes stimulated with TSA or with a combination of 5-AZA-dc and TSA revealed significant expression decline mainly for genes relevant to chondrogenesis. Based on the transcriptomic data TSA stimulation seems to be undesirable for chondrogenic differentiation of passaged cartilaginous cells.
