Nuclear pore complex (NPC), the gatekeepers of macromolecules across the nuclear envelope (NE), undergoes dynamic changes pivotal in physiology and pathology. Our study established ECM stiffness as a key driver of NB cell migration through a novel lamin A/C/E2F4/PLK1 axis, which governed post-mitotic NPC assembly and nucleocytoplasmic transport function. Targeting NPC-mediated transport may represent a promising therapeutic strategy for NB. Overall design: Analyzing whole transcriptomic profiles across different stiffness conditions aims to investigate how substrate stiffness influences gene expression and signaling pathways in neuroblastoma cells.
