Regulatory B cells are considered as a very heterogenous population of different origins and functions. We used single cell RNA sequencing (scRNA-seq) to analyze the regulatory B cells expressing granzyme B (GZMB+Bregs). Overall design: B cells from 4 healthy individuals and 6 kidney transplant recipients (3 with humoral rejection, 1 with stable function and 2 Spontaneous Operational Tolerants) were isolated from blood uning the comercialized B cell isolation kit II (Miltenyi) and separated in two conditions of non-Bregs (culture medium for 3 days) and GZMB+Bregs (culture medium supplemented with IL-21, IL-2, CpG DNA, CD40L, anti-human IgG/A/M F(ab)'2 for 3 days)
