The development of effective and targeted drug delivery systems for therapeutics and diagnostics is a challenging matter. Lipid-based nanocarriers are emerging as optimal candidates to achieve enhanced control over drug release, target selectivity and biocompatibility, thus resulting in improved pharmacokinetic profiles. The fabrication of these systems through precision microfluidics allows to obtain optimized composition, structure and surface properties with high reproducibility, by screening a large number of synthetic parameters through experimental design approaches. In this work, we synthetized optimized transethosomes as vectors for treatment of eye pathologies by screening varied lipid components and polymer coatings. The fundamental understanding of the mechanism of nanosystems interaction with membranes is essential to achieve control over intracellular drug release, correlating structural and dynamic properties to carrier functionality, thus obtaining suitable prototypes for clinical applications.
