We analyzed m6A distribution by m6A MeRIP in otder to determine the effect of PCIF1 KO over m6A distribution in human melanoma cells Overall design: mRNA from two independent clonal PCIF1 KO cell lines and one control cell line was immunoprecipitated using an antibody against m6A. RNA-Seq libraries were generated from three biological replicates for Input (RNAseq) and m6A-immunoprecipitated mRNA. Libraries were sequenced in a HiSeq with an average coverage of 20 million reads per sample. Please note that the m6A rabbit monoclonal antibody from the EpiMark N6-Methyladenosine Enrichment Kit was used. This antibody is produced by Cell Signaling and distributed by NEB in the kit, but is not sold independently and doesn't have a catalog number.
