In recent years, increasing detection of progestogens like levonorgestrel (LNG) in the environment raises concerns about their impact on fish. This study examined LNG's effects on female western mosquitofish (Gambusia affinis), assessing morphology, histopathology, and brian transcriptomics after a 30-day exposure to varying LNG levels (0, 5, 50, 500 ng/L). We found significant effects on fish body weight and growth, anal fin ratio changes, ovarian degeneration, and brain tissue abnormalities. Transcriptome analysis revealed genes linked to angiogenesis, neurotoxicity, and reproductive toxicity, shedding light on LNG's molecular impacts. Overall, LNG poses significant reproductive and neurotoxic risks to mosquitofish, offering crucial insights for environmental risk assessment of LNG.
