Replicational dilution of H3K27me3 in mammalian cells and the role of poised promoters

Differential effect of PRC2 loss on gene repression in intestinal stem cells, transit amplifying cells, and differentiated villus cells in intestine is explained by disparity in the rate of cell division and resulting difference in H3K27me3 loss. We conducted RNA-seq, ChIP-seq, scRNA-seq experiments using purified intestinal epithleal cells from wild-type and mutant adult mice and human lymphoma cell line.
