Tuberculosis (TB) remains a major global health burden, particularly in regions with a high prevalence of HIV infection, where TB-HIV co-infection accelerates disease progression, increases transmission, and promotes the emergence of multidrug-resistant Mycobacterium tuberculosis (Mtb) strains. This project aims to generate and analyze whole genome sequencing (WGS) data from clinical Mtb isolates collected from TB-HIV co-infected and non-HIV-infected patients in Pakistan to investigate genetic diversity, drug resistance mutations, virulence factors, and transmission patterns. Comparative pan genome and phylogenomic analyses will be conducted to identify core and accessory genes associated with adaptation, pathogenicity, and host immune modulation, as well as to understand evolutionary relationships and strain distribution. The study will also support computational drug discovery by identifying novel therapeutic targets and evaluating potential inhibitors through molecular docking and virtual screening. Overall, the generated genomic data will enhance TB surveillance and contribute to improved diagnostics and therapeutic strategies for TB-HIV co-infection.
