Purpose: The goals of this study are to elucidate the underlying mechanism for the regulation of HSC divisions. Methods: Before or after wild type (WT) mice were treated with 5-fluorouracil (5-FU), 15000 cells of HSCs were sorted. These cells were subjected to ATAC-seq using Next-seq. The sequence reads that passed quality filters were analyzed by FASTX-toolkit. After 6 or 10 days from 5-FU administration, HSCs (CD150+ CD48- EPCR+ Lineage-) were sorted were sorted, and subsequently ATAC peaks were generated by deep sequencing using Next-seq.
