We report polyadenylated transcripts associated with R521C mutant FUS in the insoluble fraction. Overall design: FLAG-tagged R521C mutant FUS was transiently expressed in U87-MG glioma cells for 36 hours followed by NP-40 lysis. The NP-40 insoluble fraction was extracted with 0.1M urea and the supernatant was used for RNA-IP using FLAG antibody. RNAs co-IPed with FLAG-tagged mutant FUS were subjected to 3' region extraction and deep sequencing (3'READS).
