Somatic cell nuclear transfer (SCNT) is an irreplaceable tool to understand nuclear reprogramming and totipotency, it also facilitated the generation of genetic modified and gene editing animals for various applications. However, with mice as the only exception, SCNT has remained an inefficiency process for many years. It was reported that the developmental potential of early-cleaving SCNT embryos is higher than that of later-cleaving embryos, yet the underlying regulation mechanism remains largely unknown. To better understand the molecular mechanisms of these changes, single cell RNA-seq of pig SCNT embryos which are early-cleaving (24 h post-activation) and late-cleaving (36 h post-activation) were performed.
