- Background: Optimized diets during the first period of life may be most effective for improving gut and overall health. Here, we set up an interdisciplinary research pipeline to evaluate gut health benefits of early life nutrition ingredients through advanced integration of in vitro and modeling technologies that represent the infant gut environment. - Methods: In our InTESTine platform (TM), biopts of piglet gut tissue (Jejunum or Colon) were exposed (0-6h) to various infant/piglet-formula-milk based food matrices (-/+ prebiotics; -/+ predigestion by infant/piglet fecal microbiome). RNA expression of the piglet gut tissue biopts was measured by RNAseq. - Results: To be published in various papers. - Conclusion: To be published in various papers. RNAseq expression of piglet (age 3wk) gut tissue biopts (J=jejunum or C=colon) after 0-6h exposure to various infant/piglet formula-milk based food matrices (IFM = 10% Infant Formula Milk, PMR = 10% Piglet Milk Replacer) -/+ addition of prebiotics (2'FL=2'-fucosyllactose, LGG = Lactobacillus rhamnosis Goldin Gorbach, GOS = Galacto-oligosaccharide, ZnO = Zinc oxide) and -/+ predigestion of the milk-based foodmatrices (-/+ prebiotics) by microbiome from either breast/formula-fed human infants (BfH = microbiome Brest-fed Human infants, FFH = microbiome Formula-fed Human infants) or healty/dysbiotic piglets (DysP = microbiome dysbiotic piglets, HeaP = microbiome healthy piglets) in our i-screen platform (TM; i = i-screen = predigested foodmatrix -/+ prebiotic). 0 = no added: prebiotic, food matrix or microbiome. SIEM = blanc microbiome medium of the i-screen platform (no addiditons of prebiotic, food matrix and microbiome).
