Whole-genome pooled sgRNA screen using immortalize bone marrow-derived mouse macrophages with Cas9 and an NFKB-GFP reporter system (iBMDM-NFKB-Cas9) The goal of this experiment was to 1) identify regulators of NFKB signaling 2) identify genes important for viability iBMDM-NFKB-Cas9 cells were infected with a custom whole-genome sgRNA library and were stimulated with 200ng/ml LPS and FACS sorted for collect top/bottom 20% of cells. For the viability screen, cells were collected at day 0 and day20.
