Description: This dataset contains all raw data supporting the findings of the study "Baicalin-Geniposide Ameliorates Neurological Injury and Secondary Acute Lung Injury Induced by Cerebral Ischemia Reperfusion by Blocking Nucleoplasmic Transfer of HMGB1." Study summary: Baicalin-Geniposide (BG), a natural compound combination, was investigated for its protective effects against cerebral ischemia-reperfusion injury and secondary acute lung injury (ALI). Using LPS-stimulated BV2 microglia and RAW264.7 macrophage co-cultures, we demonstrated that BG dose-dependently suppressed pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) and nitric oxide (NO), while attenuating HMGB1 nucleocytoplasmic translocation via JAK2/STAT3 inhibition. In MCAO/R rats, BG (25–50 mg/kg) reduced cerebral infarct volume, neurological deficits, and lung edema. Mechanistically, BG blocked HMGB1 nuclear export in ischemic brains, decreasing HMGB1 levels in serum and lungs. Data contents: 01_in_vitro_data: Raw data from BV2 and RAW264.7 cell experiments, including CCK-8 cell viability, ELISA (TNF-α, IL-1β, IL-6), and NO assays. 02_in_vivo_data: Raw data from MCAO/R rat experiments, including neurological scores, infarct volume (TTC staining), lung wet/dry weight ratio, tissue ELISA (brain and lung), and serum HMGB1 levels. 03_histology: Original histopathological images (HE staining of brain and lung tissues) and immunofluorescence images (HMGB1 in brain tissues and BV2 cells). 04_western_blot: Original full Western blot membrane images for JAK2/STAT3/HMGB1 pathway analysis in BV2 cells and brain tissues, including JAK2 activator (CA1) and inhibitor (AZD1480) experiments. All data are organized by figure and experiment type. Western blot images include molecular weight markers and lane labels. ELISA data include raw OD values and standard curves.
