Eukaryotic species generally have multiple chromosomes. Here we have created a functional single chromosome yeast from a Saccharomyces cerevisae haploid cell containing sixteen linear chromosomes through successive chromosome end-to-end fusions and centromere deletions. In order to detect the configuration change due to fusion events, We carried out the chromosome conformation capture (3C)-derived Hi-C assays on BY4742 and SY14 cells, as well as two intermediate strains SY6 (containing nine chromosomes including seven fused and two native chromosomes) and SY13 (containing only two fused large chromosomes) using MboI enzyme.
