We used mircoarray to identify the potential regulation on the gene level between DDIT4+/+ and DDIT4−/− cells treated with 5-FU at 24h. Genes related to protein secretion, androgen response, mitotic spindle, hypoxia, TNFa signaling via NFKB, apoptosis, glycolysis, mTORC1 signaling, G2M checkpoint, KRAS signaling up are enriched in the DDIT4KO gastric cancer cell treated with 5-FU. RNA from gastric cancer cells was isolated from DDIT4KO and NC MGC-803 cells that were treated with 5-FU 24 hours. DDIT4KO cells treated with 5-FU have significantly increased proliferation compared with NC cells. DDIT4KO and NC MGC-803 gastric cancer cells treated with 5-FU was compared to determine DDIT4 tragets under stimulation in gastric cancer.
