We generated mRNAseq data to assemble transcriptomes for Aloe arborescens Mill., Aloe buettneri A.Berger, Aloe vera (L.) Burm.f. and Aloidendron barberae (Dyer) Klopper & Gideon F.Sm. We sampled one plant accession for each of the four species from the curated living collections at Royal Botanic Gardens, Kew. Leaves were sampled in the greenhouse and immediately frozen in dry ice. Three technical replicates were generated for each biological sample. RNA was isolated from frozen material using a Qiagen Plant RNEasy kit. mRNA was purified from rRNA and tRNA material using a polyA-tail column cleaning procotol. cDNA libraries were built using Illumina TruSeq kits and sequenced on an Illumina NextSeq machine. Sequence reads were assembled into transcript contigs using Trinity. The technical replicate with the highest Trinity "gene" count was kept for the design of the Alooideae target capture probe set.
