Research hypothesis: Icariin ameliorates cisplatin-induced premature ovarian insufficiency (POI) by inhibiting ferroptosis in human ovarian granulosa cells (KGN) via activation of the Nrf2/GPX4 signaling pathway. What the data show: The dataset includes raw and processed data from three categories: Network pharmacology & molecular docking – predicted targets of icariin against POI, PPI network hub genes, GO/KEGG enrichment results, and docking scores/binding conformations of icariin with key targets (e.g., NFE2L2, NOX4, PTGS2). In vitro experimental data – from cisplatin‑treated KGN cells with or without icariin (5–20 µM) or the Nrf2 inhibitor ML385, including: Cell viability (CCK‑8) Ferroptosis markers: intracellular Fe²⁺, MDA, GSH, LDH, ROS (DCFH‑DA), lipid peroxidation (BODIPY 581/591 C11) Protein expression (Western blot): Nrf2, HO‑1, GPX4, ACSL4 mRNA expression (qRT‑PCR): Nrf2, HO‑1, GPX4, ACSL4 Statistical summaries – mean ± SD, number of replicates (n=3–4 per group), and significance indicators (P values, ANOVA with post‑hoc tests). Significant findings: Cisplatin significantly reduced KGN cell viability, increased Fe²⁺, MDA, LDH, ROS and lipid peroxidation, depleted GSH, downregulated Nrf2/HO‑1/GPX4, and upregulated ACSL4. Icariin (20 µM) reversed all these effects in a dose‑dependent manner. The protective effects of icariin were abolished by the Nrf2 inhibitor ML385, confirming pathway dependence. How to interpret and use the data: Blot images and qPCR raw data can be used to verify band densities and CT values. Ferroptosis marker values (Fe²⁺, MDA, GSH, LDH, ROS, BODIPY) reflect the degree of lipid peroxidation and oxidative stress. Comparisons between groups (Ctrl, Cis, Cis+ICA, Cis+ML385, Cis+ML385+ICA) allow assessment of icariin’s protective effect and Nrf2 dependency. Network pharmacology files (target lists, enrichment tables, docking scores) can be reused for comparative or meta‑analyses of natural products against POI. All raw data (blots, spreadsheets, docking outputs) are organized by figure number for easy cross‑reference with the published article.
