Yeast rho- mutants

Whole genome sequencing of Saccharomyces cerevisiae strains (derivatives of W303 genetic background) carrying deletions in mtDNA (rho- genotype) using Illumina sequencing platform. We sequenced 17 independently obtained mutant strains each derived from the same parental strain W303-1B (ade2-101 his3-11 trp1-1 can1-100 leu2-3,112, GAL, psi+, URA3 MATalpha) carrying different deletions in their mitochondrial genome. For two strains, we also sequenced five subclones (amplified from single colonies) to analyse secondary mutations in mtDNA. Additionally, we sequenced the strain with wild-type mtDNA sequence (rho+ genotype) and two strains completely lacking mtDNA (rho0 genotype).Strains were cultivated on reach medium (YPD = 1% yeast extract, 2% bacto-peptone, 2% glucose).
