Background & purpose:This dataset supports the findings of our study titled "Steroid biosynthesis pathway counteracts iron overload‑induced ferroptosis in mouse granulosa cells". The research investigates how iron overload impairs female reproductive function and reveals a protective role of the steroid biosynthesis pathway against ferroptosis in ovarian granulosa cells. Data content – five folders: Mouse – in vivo data from the iron‑overload mouse model (ferric citrate gavage): ovarian iron content, estrous cycle, pregnancy rates, litter sizes, serum estradiol, ovarian histology (H&E). Metabolomics – untargeted LC‑MS metabolomics of freshly isolated mouse granulosa cells (raw and processed data). Transcriptomics – RNA‑seq of granulosa cells (gene expression profiles, counts, differential expression). KK-1 cell data – in vitro experiments in KK1 cells under iron overload (2 mM ferric citrate ± deferoxamine): Fe²⁺, ROS, GSH, NADPH/NADP⁺, MDA, lipid peroxidation imaging, trypan blue, qRT‑PCR. KK-1 cell Sreb2 knockdown... – siRNA knockdown of Srebf2: efficiency, downstream gene expression, lipid peroxidation and trypan blue after knockdown under iron overload. Methods & processing:Detailed protocols are in the manuscript. Briefly: C57BL/6J female mice (ferric citrate 120 mg/kg/day for 40 days); LC‑MS metabolomics (Metware Biotechnology Co., Ltd., Wuhan); RNA‑seq (Metware Biotechnology Co., Ltd., Wuhan); KK1 cell culture, 2 mM FC ± 100 μM DFO, siRNA; assays (ELISA, flow cytometry, fluorescence microscopy, qRT‑PCR).
