Salivary exosomes were collected from healthy people and patients with benign and malignant tumors, and small RNA sequencing was performed to screen differential small Rnas for tumor screening We collected saliva from 4 healthy volunteers, 4 patients with benign ovarian tumors and 4 patients with malignant ovarian tumors. Exosomes were isolated by ultracentricentrifugation and identified by electron microscopy and particle size. RNA was extracted by abs60263 exosome RNA extraction kit. RNA purity (OD260/280,OD260/230 ratio) was detected by NanoDrop™ One/OneC. The Qubit™ RNA HS Assay Kit was used for accurate quantification using a Life Invitrogen Qubit® 3.0 fluorescence quantizer. Kapa qPCR was used to quantify the library concentration accurately. The Agilent 4200 TapeStation system was used: RNA integrity (RIN value) and library fragment size were precisely determined.The clustering of the index-coded samples was performed on a cBot Cluster Generation System using TruSeq PE Cluster Kit v3-cBot-HS (Illumia) according to the manufacturer’s instructions. After cluster generation, the library preparations were sequenced on an Illumina platform.
