In this experiment the relationship between SOX2 levels and WNT-induced gene expression was assayed using RNA-seq during embryonic stem cell differentiation. A doxycycline inducible cell line (SOX2 TetON) was employed to experimentally control SOX2 expression. Overall design: Transcriptional profiles were generated from three independent biological replicates in four experimental conditions. Sox2 TetON ES cells differentiated in the presence or absence of 1ug/ml doxycycline were compared to differentiated WT epiblast progenitor cells cultured in the presence or absence of the WNT agonist CHIR99021.
