The goal of this project was to determine the effect of N-acetylcysteine in drinking water on progression of leukemia in a mouse model initiated by deletion of PU.1 and Spi-B. Mice that delete PU.1 and Spi-B in the B cell lineage (Mb1-CreDPB mice) develop precursor B cell acute lymphoblastic leukemia with a median time to euthanasia of 18 weeks. The pre-B-ALLs that develop in this model have a mutational signature characteristic of 8-oxoguanine DNA damage induced by reactive oxygen species. Mb1-CreDPB mice were placed into two groups, one on regular drinking water and one on drinking water containing 1g/L of the antioxidant N-acetylcysteine. We hypothesized that N-acetylcysteine would alter gene expression in leukemias developing in these mice. Total RNA was prepared from leukemias from 8 mice on regular drinking water and 8 mice on N-acetylcysteine (NAC) drinking water. This is a total of 16 biosamples. For each biosample, libraries were prepared using Truseq Stranded Total RNA Library Prep Kit v2 (Illumina).Paired-end Illumina RNA-sequencing was performed using the Illumina HiSeq4000 by Genome Quebec. Each biosample has two separate files containing read 1 and read 2. Raw data in gzipped fastq format is provided.
