E. coli RNA Deep Sequencing. Escherichia coli K-12

This sequencing was carried out to assess how sequencing depth affects profiling of bacterial transcriptomes by RNA-Seq. Total RNA was isolated from a single E. coli K-12 LB culture in mid-log growth (OD600 ~ .500). Illumina libraries were constructed from this RNA with and without depletion of ribosomal RNA using the RiboZero Gram negative kit (Epicenter). Each library was sequenced in a single lane of Illumina HiSeq 2000 to yield 101 paired end reads.
