Chromatin immunoprecipitation DNA-sequencing (ChIP-seq) for Pol ? in HEK293T cells. Overall design: Genome editing-based integration of three tandem repeats of the FLAG epitope tag (3ÃFLAG tag) into endogenous Pol ? was conducted. Then we crosslinked DNA and proteins in chromatin, fragmented DNA, immunoprecipitated endogenous Pol ? and its associated genomic DNA using anti-FLAG M2 beads, and subjected the resulting DNA fragments for next-generation sequencing analysis.
