Botryococcus race B produces methylated triterpenoids which are potentially useful as biofuels. The project involved selectively assembling the orthologue and all paralogues of squalene synthase gene sequences, using Illumina data with deep (>50x) coverage, including single end reads and paired end reads. The assembly technique was based on SOAPdenovo v 1.04, with some manual curation using Sequencher for the particular contigs of interest. Paired end libraries of 2 kb and 4kb respectively were prepared by- and sequenced by- Illumina Hayward to enable crossing of introns, and hence assembly of contiguous full exons. The work was submitted to Frontiers in Microbiology, as Moore et al. 2021, and has evolutionary aims to do with assessing the order of gene duplications.
