Microcosms were established as follows: Mason jars were filled with 10 g of Wyoming MX-80 bentonite mixed with 3 mL of sterile Type I water (to achieve a water activity of 0.99), mixed thoroughly using one of three similar methods, and incubated for one week at 45 degrees C under oxic conditions. After incubation, aerobic heterotrophs were cultured. DNA was extracted from bentonite and from culture biomass and sequenced.
