Lipid nanocapsules (LNCs) are redefining the boundary between nanocarriers and active drugs. Traditionally regarded as inert delivery systems, LNCs are now emerging as bioactive nanostructures capable of engaging molecular signaling pathways. Composed of medium-chain triglycerides (MCTs) and surfactants, these nanosystems display intrinsic metabolic activity that extends far beyond cargo transport. Here, we demonstrate that blank LNCs act as synthetic lipid agonists of G protein-coupled receptors (GPCRs), reproducing the pharmacological profile of glucagon-like peptide-1 receptor agonists (GLP-1RAs), a class of antidiabetic drugs that stimulate insulin secretion.
