Jaridon 6, an ent-kaurene diterpenoid derivative from Rabdosia rubescens (Hemsl.) Hara, possesses strong antitumor activity in esophageal cancer cells. This study explored the underlying molecular events of Jaridon 6’s anti-tumor activity in esophageal cancer cells through the cDNA microarray. Overall design: We wanted to obtain early changes of RNA by Jaridon 6's action on the esophageal cancer 109 cells, especially in early apoptosis stage. The IC50 of Jaridon 6 is 15 µM. When the esophageal cancer 109 cells were acted by 15 µM Jaridon 6 for 16h, the apoptosis rate is approximately 40%, near to the early apoptosis stage. So the esophageal cancer 109 cells were treated with 15 µM Jaridon 6 for 16 h for RNA extraction and hybridization on Affymetrix microarrays.
