Downsampled images for FijiFISH tutorial.The file folder contains the original z-stacks for analysis in FijiFISH. Once you have run all steps, the analyzedTables folder is where one can begin the RUHi portion of analysis. See our other figshare folder with the CSVs contained in the analyzedTables folder to start straight away with the RUHi tutorial.Data contains 4 rounds of imaging of mouse 20 micron thick mouse brain sections with DAPI as a marker for registration and segmentation. First, animals underwent stereotaxic surgery wherein retrograde viral tracers were injected into the retrosplenial cortex (RSC) and lateral entorhinal cortex (LEC). After allowing these tracers to express, the animals were perfused, sectioned, stained with DAPI, and the circuit mapping was imaged on a Leica SP8 WLL confocal. After this, the tissue was decoverslipped and underwent RNAscope HiPlex, which was then imaged using identical settings to the viral imaging over the course of 3 rounds of imaging.This data was originally published by Erwin*, Bristow*, et al (<i>eLife</i> 2023) DOI: 10.7554/eLife.68967
