This record contains the raw single-cell microscopy images and the numerical source data underlying the figures of Dyer et al., "Chaperone condensates buffer the heat shock response against pleiotropic inputs." Raw microscopy images (spinning-disk confocal; Slidebook, TIFF) are organized by imaged protein / experiment: images_Rpl26a.zip — newly synthesized Rpl26a-Halo vs. nucleolar marker Nsr1 (orphan ribosomal protein accumulation). images_Hsf1.zip — Hsf1 condensation across stresses. images_Ssa1_Hsp70.zip — Hsp70 (Ssa1) condensation. images_Med15.zip — Mediator subunit Med15 condensation. images_Pol_II.zip — RNA Pol II (Rpb3) condensation. images_Sis1_FRET.zip — Sis1-mEos3.2 DAmFRET biosensor of Sis1 self-association. images_AA_imaging.zip — Sis1 anchor-away (nuclear Sis1 depletion) colocalization imaging. Figure source data (source_data_prism.zip): GraphPad Prism files containing the quantified data plotted in the figures (condensation indices, colocalization, FRET, RNA-seq volcano/PCA of HSR genes). Stress abbreviations used throughout: NT/control = unstressed; HS = heat shock; Ars = sodium arsenite; AZC = azetidine-2-carboxylic acid; DZA = diazaborine; NaCl = hyperosmotic salt; CHX = cycloheximide (translation block); 1,6-HD = 1,6-hexanediol; AA = Sis1 anchor-away (rapamycin-induced nuclear depletion).
