Through a genome-wide CRISPR/Cas9 screen in NSCLC cell lines, we discover that SMARCA4 knockout significantly protects tumour cells from ferroptotic cell death. ATAC-Seq and CUT&TAG Seq suggest that ALDH16A1 is a potential target of SMARCA4. Together, in this work, we uncover a critical mechanism by which SMARCA4 deficiency enables NSCLC cells to evade ferroptosis, thereby rationalizing the targeting of the SMARCA4/ALDH16A1/TXN/ferroptosis axis as a promising therapeutic approach for patients with SMARCA4-deficient NSCLC.
