RNA-seq based transcriptome analysis of human fibroblast, primed and naïve induced pluripoent stem cells Three different fibroblast lines were reprogrammed to naïve pluripotency by transfection of modified messanger RNAs in microfluidics. Eight niPSC lines have been generated and expanded under standard conditions before profiling by RNAseq. An isogenic primed iPSC line (HPD00) has been also generated and analysed, as well as a naive PSC line (H9 Reset), obtained by expression of NANOG-KLF2 transgenes in H9 primed hESCs.
