Transcriptomic profiling of MLL-r AML PDX cells treated with CDKI-73 and/or JQ1 RNA was extracted from MLL-r AML PDX cells treated with CDKI-73, JQ1, or a combination of aforemetioned compounds for 4 hours, and transcriptome was profiled using RNA-Seq Cryopreserved MLL-r AML (AML-18) PDX cells were thawed and allowed to equilibrate for 3 hours prior to in vitro drug treatment with 250 nM CDKI-73, 1480 nM JQ1 the combination of both drugs or vehicle control (DMSO). After 4 hours, total RNA was extracted using TRIzol (Invitrogen) and purified using the RNeasy kit (Qiagen). Whole transcriptome libraries were prepared using the Illumina TruSeq Stranded Total RNA Sample Preparation Kit with Ribo-Zero Gold rRNA depletion. Sequencing was performed using the Illumina HiSEQ2500 platform as 100 bp single ended sequencing
