We use RNAseq to analyze the transcriptomes of adjacent tissue sections from the same melenoma, one in which PTEN protein expression is retained and one in which it is lost. This direct, intratumoral comparison provides a syngeneic background on which to assess tumor-intrinsic and -extrinsic effects of PTEN loss including cancer cell proliferation and immune cell infiltration.
