Cortical single-cell RNA sequencing of the microglia specific TREM2 inducible mouse models

We induced TREM2 expression at 2 months of age in our TREM2 mouse models and harvested the animals at 4 months of age. The single cells from the cortex were isolated and subjected to the single-cell RNA seq to investigate the effects of TREM2 on the transcriptomic profiles at single cell level in healthy condition. Our data revealed that TREM2-WT expression triggered the downstream mTOR/EIF2 signaling pathways and activated microglia by elevating Apoe expression; whereas TREM2-R47H had no effects on mTOR/EIF2 signaling pathway and minor impact on activating microglia antigen presentation pathway. Four mice (2 males and 2 females) were analyzed for each of the four groups, including TREM2-WT-Ctrl, TREM2-WT-Induced, TREM2-R47H-Ctrl, and TREM2-R47H-Induced.
