Gene expression profile at single cell level of GFP labeled islet ß cells from streptozotocin (STZ) treated Ins/ß-lineage-labeled mice

Islet ß cells suffer from de- and trans-differentiation after injury. We used single cell RNA sequencing (scRNA-seq) to analyze the diversity of ß cells in from STZ-induced diabetes. Overall design: Ins/ß-lineage-labeled (Ins1-CreERT; Rosa26-mT/mG) mice underwent 5-day STZ treatment after 5-day tamoxifen (TAM) treatment and 2-wk washout. At 12wk after STZ treatment, islets were isolated and dispersed into single cells. GFP positive cells were isolated by Fluorescence-activated cell sorting (FACS) and analyzed using scRNAseq.
