Using normal healthy wild-type mice (normal) and transgenic BCR-ABL mouse model (CML), we identified differences in gene expression between various normal and CML skeletal stem cell subsets. Briefly, we identified that the bone-forming progenitors (Thy) and stroma-forming progenitors (6C3) were significantly expanded upon leukemia development due to increased cycling. Furthermore, we observed significant perturbation of their capacity to support normal hematopoietic stem cell function (HSC). Interestingly, we observed that CML 6C3 cells better support leukemic stem cell function. Therefore, normal and CML mSSC subsets were FACS sorted and their global transcriptome was analyzed using RNA-Seq.
