The effects of knockout and overexpression of CaRAP2.6L gene on salt tolerance of pepper seedlings

In this study, the pepper variety 'Xiangla 762' (XL762) was used as the material. Uniform seedlings were obtained through seed disinfection, germination promotion and seedling raising. The root-inducing Agrobacterium-mediated genetic transformation technology was utilized to construct and screen out seedling materials with clear genetic background of root system, including empty vector, CaRAP2.6L gene knockout and overexpression lines. After the gene expression levels were verified by qRT-PCR, they were used for subsequent experiments. When the seedlings grew to five leaves and one heart, they were transplanted into the hydroponic system for cultivation. The experiment was set up with a control group (without salt) and a salt treatment group (150 mM NaCl), with three replicates in each group and six plants in each replicate. Molecular verification and screening of positive materials were completed before the start of the treatment.
