RNA-Seq of rat cardiac fibroblasts depleted for Yap and Wwtr1

Primary neonatal rat cardiac fibroblasts were isolated and subjected to siRNA mediated Yap and/or Wwtr1 knockdown Overall design: Primary cardiac myocytes were isolated from two-day old rats via neonatal heart dissociation kit (Miltenyi Biotec) according to the manufacturer's protocol followed by percoll gradient separation. Cells were transfected with siRNAs designed against Yap, Wwtr1, a combination of Yap and Wwtr1, or universal negative control siRNAs. Transfected cells were collected 48 hours post transfection for RNAseq.
