Datasets for "A 3D printed scaffold-coupled perfusion bioreactor promotes uniform cell distribution and osteogenic differentiation in serum-free conditions"
收藏资源简介:
This dataset contains the quantitative data, statistical analyses and RNA sequencing differential expression outputs supporting a study of scaffold coupled perfusion culture for human bone tissue engineering, as a supporting dataset for a study titled "A 3D printed scaffold-coupled perfusion bioreactor promotes uniform cell distribution and osteogenic differentiation in serum-free conditions". Primary human bone marrow derived mesenchymal stromal cells from three donors were seeded onto 3D printed porous polylactic acid scaffolds and cultured in the BioAxFlow bioreactor under chemically defined, serum free osteogenic conditions. Following dynamic seeding, constructs were maintained for 14 days under static conditions or perfusion at total flow rates of 40 or 80 mL/min. The study combined scaffold characterization, computational fluid dynamics, cellular imaging, mineralization assessment and bulk RNA sequencing to investigate spatial tissue development and cellular responses to perfusion. The deposited files include quantitative source data underlying the manuscript figures, statistical analyses of the measured outcomes and DESeq2 differential expression results for comparisons between culture conditions. These data support the reported improvements in internal cellular distribution and mineralization under perfusion, together with differences in hypoxia associated responses, extracellular matrix organization, adhesion and osteogenic regulation. The deposition supports independent assessment and reuse of the quantitative findings and differential expression results reported in the associated manuscript.
