Rhopalosiphum padi whole-body diurnal transcriptome. Rhopalosiphum padi

The goal for this project was to characterize the diurnal transcriptome of adult Rhopalosiphum padi whole bodies. RNA was sampled from an age synchronized colony every four hours at six timepoints over a 24h period. Plants and aphid colonies were maintained under a short-day photoperiod (12L:12D). Three replicates of thirty aphids were removed from the colony and immediately frozen in liquid nitrogen at each timepoint. Total RNA was extracted with the Quick-RNA Miniprep Kit (Zymo Research, Irvine, CA). RNA samples were sent to Novogene Corporation Inc. (Sacramento, CA) for mRNA enriched library preparation and paired-end RNA sequencing on the Illumina HiSeq 2000 platform.
