We analysed the transcriptional signature of bone marrow Nestin+ mesenchymal stromal cells extracted from the bone marrow of mice engrafted with human AML cell lines and compared it to the one of untransplanted mice Overall design: Immunodeficient mice were transplanted with human AML cell lines, human AML cells derived from patients (blood or bone marrow), or with normal human hematopoietic cells (cd34+) derived from cord blood. Mice were sacrificed once assessed the bone marrow engraftment, and the bones were processed to isolate niche cells using the depicted marker. RNA was extracted, sequencing libraries were prepared and sequenced. "early" stands for isolated at early time points of the leukemic progression "late" stands for isolated at late time points of the leukemic progression "ut" stands for untreated "ch" stands for cherry, the marker we used to isolate the leukemia proximal niche.
