Velcrins are molecular glues that kill cells by inducing the formation of a proteincomplex between the RNAse SLFN12 and the phosphodiesterase PDE3A. Formation of thecomplex activates SLFN12, which cleaves tRNA Leu (TAA) and induces apoptosis. Velcrins suchas the clinical investigational compound, BAY 2666605, were found to have activity acrossmultiple solid tumors cell lines from the Cancer Cell Line Encyclopedia (CCLE), includingglioblastoma cell lines. In this study, glioblastoma cell lines that are either sensitive or resistant to BAY 2666605 were treated for 24 hours and gene expression changes were measured by RNAseq.
